Cultivation of Insect Cells in Airlift Reactors: Influence of Reactor Configuration and Superficial Gas Velocity

چکیده
Large-scale cultivation is an essential step towards the feasible production of baculovirus in insect cell cultures. Airlift reactors appear to offer considerable advantages over other insect cell culture systems. In order to evaluate the impact of reactor design on the behavior of insect cell cultures, the IPLB-Sf-21 cell line was cultivated in three different concentric tube airlift reactors that differ in their geometrical parameters. The ratio of downcomer to riser cross sectional areas, the shape of the bottom and the ratio of height to diameter of the reactor proved to be important since them produce significant differences on cell growth behavior. Modifying the reactor design the cellular growth rate could be improved from 0.016/h to 0.031/h, while the maximum viable cell density could be elevated from 9x10 to 2.4x10 cells/ml. Once selected a reactor configuration, the influence of gas flow rate was determined, finding an optimal value of superficial gas velocity that renders sufficient oxygenation without any significant effect on the cellular viability. In addition, the influence of the reactor design on fluid circulation in the reactor was tested.

Hydrodynamics and mass transfer inthree-phase airlift reactors for activated Carbon and sludge filtration
A bioreactor refers to any manufactured or engineered device that supports a biologically active environment. These kinds of reactors are designed to treat wastewater treatment. Volumetric mass transfer coefficient and the effect of superficial gas velocity, as the most important operational factor on hydrodynamics, in three-phase airlift reactors are investigated in this study. The experiments...
متن کاملThe Effects of Geometry and Operational Conditions on Gas Holdup, Liquid Circulation and Mass Transfer in an Airlift Reactor
In airlift reactors transport phenomena are achieved by pneumatic agitation and circulation occurs in a defined cyclic pattern through a loop. In the present work, the effect of geometrical relations on gas holdup and liquid velocity, and consequently on the gas-liquid mass transfer coefficient, was studied in a 6liter airlift bioreactor with AD/AR = 0.63; AD, downcomer cross-sectional area, an...
متن کاملGas holdup and mixing characteristics of a novel forced circulation loop reactor
Gas holdup and mixing time were characterized in a forced circulation internal loop draft-tube reactor (unaerated aspect ratio≈ 6, downcomero-riser cross-sectional area ratio = 0.493) as functions of the forced liquid superficial velocity in the riser (ULr) and the gas superficial velocity in he riser (UGr). Data were obtained in air–water system. The operation ranges were 0≤ULr ≤ 0.051 m/s and...
متن کاملAxial inhomogeneities in steady-state dissolved oxygen in airlift bioreactors: predictive models
Models were developed for prediction and interpretation of the observed steady-state axial dissolved oxygen concentration profiles in tall airlift bioreactors. The observed concentration profiles were non-linear because of a combination of hydrodynamic and mass transport factors. The profiles were influenced mainly by the liquid-phase axial dispersion coefficient, the volumetric overall gas–liq...

For an integrated nitrification-denitrification processes, nitrite formation in the aerobic stage leads to big savings. Recently experimental observations (Garrido et al., 1996) have shown that it is possible to obtain full ammonium conversion with approximately 50 % nitrate and 50 % nitrite in the effluent of a biofilm airlift suspension reactor. With oxygen concentrations between 1 and 2 mg/l...
